CASP7(Caspase-7)是半胱天冬酶(caspase)家族的重要成员,属于执行型caspase(executioner caspase),主要参与细胞凋亡(程序性细胞死亡)的最终执行阶段。其生物学功能是通过切割特定蛋白质(如PARP、DFF45等)来触发细胞凋亡的形态学和生化变化,如DNA断裂和细胞膜起泡。CASP7通常以无活性的酶原形式存在,被上游caspase(如caspase-3或caspase-8)激活后形成活性异源二聚体。主要作用位点包括细胞质和细胞核,尤其在免疫细胞的凋亡调控中起关键作用。突变可能导致CASP7功能丧失,抑制细胞凋亡,与癌症(如结直肠癌、乳腺癌)的发生相关;某些突变也可能增强其活性,导致过度凋亡,与神经退行性疾病(如阿尔茨海默病)有关。CASP7过表达会加速细胞凋亡,可能引发组织损伤或自身免疫疾病;而表达降低则可能导致凋亡受阻,促进肿瘤发展和化疗耐药。它属于caspase基因家族,该家族成员均含半胱氨酸蛋白酶结构域,依赖天冬氨酸特异性切割底物,分为启动型(如caspase-8/9)和执行型(如caspase-3/6/7),共同调控炎症和凋亡。CASP7与caspase-3功能高度重叠,但敲除实验显示其对某些凋亡信号(如死亡受体通路)更敏感。目前中文术语“半胱天冬酶”已广泛接受(原英文caspase,源自cysteine-dependent aspartate-specific protease)。

This gene encodes a member of the cysteine-aspartic acid protease (caspase) family. Sequential activation of caspases plays a central role in the execution-phase of cell apoptosis. Caspases exist as inactive proenzymes which undergo proteolytic processing at conserved aspartic residues to produce two subunits, large and small, that dimerize to form the active enzyme. The precursor of the encoded protein is cleaved by caspase 3 and 10, is activated upon cell death stimuli and induces apoptosis. Alternatively spliced transcript variants encoding multiple isoforms have been observed for this gene. [provided by RefSeq, May 2012]
