CCL19(Chemokine (C-C motif) ligand 19,趋化因子C-C基序配体19)是一种由趋化因子基因家族编码的小分子分泌蛋白,属于CC亚家族(β-趋化因子)。该家族成员均含有两个相邻的半胱氨酸(C-C结构),主要功能是通过与G蛋白偶联受体结合介导免疫细胞迁移。CCL19主要由淋巴器官(如淋巴结、脾脏)中的基质细胞和树突状细胞分泌,其受体为CCR7(趋化因子受体7)。CCL19的核心生物学功能是引导T细胞、树突状细胞和B细胞向次级淋巴器官定向迁移,从而调控适应性免疫应答的启动。其作用位点集中在淋巴组织的T细胞区(如淋巴结副皮质区),通过形成浓度梯度吸引表达CCR7的免疫细胞归巢。CCL19基因突变可能导致其蛋白结构异常,表现为免疫细胞迁移障碍,与某些原发性免疫缺陷病相关。该基因过表达常见于类风湿性关节炎、多发性硬化等自身免疫疾病,会加剧炎症部位淋巴细胞浸润;而表达降低则可能影响疫苗免疫效果或肿瘤免疫监视功能。在肿瘤微环境中,CCL19表达下降可能导致树突状细胞无法有效激活T细胞,促进免疫逃逸。值得注意的是,CCL19与同家族成员CCL21共享CCR7受体,但两者在组织分布和结合亲和力上存在差异。该基因家族(CC趋化因子家族)的共性包括:分子量8-10kDa、依赖二硫键维持空间结构、通过趋化作用调控白细胞运输。目前研究发现CCL19还具有非趋化功能,如促进树突状细胞成熟和调节淋巴管生成。在临床应用中,重组CCL19被探索作为肿瘤免疫治疗的佐剂,用于增强抗肿瘤T细胞反应。

This antimicrobial gene is one of several CC cytokine genes clustered on the p-arm of chromosome 9. Cytokines are a family of secreted proteins involved in immunoregulatory and inflammatory processes. The CC cytokines are proteins characterized by two adjacent cysteines. The cytokine encoded by this gene may play a role in normal lymphocyte recirculation and homing. It also plays an important role in trafficking of T cells in thymus, and in T cell and B cell migration to secondary lymphoid organs. It specifically binds to chemokine receptor CCR7.
