EZH1(Enhancer of Zeste Homolog 1)是果蝇Zeste基因增强子的人类同源基因之一,属于多梳抑制复合物2(PRC2)的核心成员,与EZH2共同组成Polycomb基因家族。该家族以表观遗传调控著称,通过催化组蛋白H3第27位赖氨酸的三甲基化(H3K27me3)来抑制靶基因表达,从而在细胞分化、发育和稳态维持中发挥关键作用。EZH1与EZH2功能部分重叠,但EZH1主要存在于非增殖或终末分化细胞中,其酶活性较低且依赖PRC2复合物的特定亚型(含SUZ12和RbAp46/48)。EZH1的突变可能导致其甲基转移酶功能异常,进而影响造血干细胞分化、肌肉再生等过程,与骨髓增生异常综合征(MDS)和某些肌病相关。过表达EZH1可能增强基因沉默效应,导致分化阻滞,例如在白血病中与EZH2协同促进肿瘤发生;而敲低EZH1会削弱PRC2的冗余调控功能,尤其在EZH2缺失时引发胚胎致死或组织再生障碍。值得注意的是,EZH1在心脏和骨骼肌中高表达,其下调可能导致肌细胞分化异常。Polycomb家族基因的共性包括:通过染色质修饰调控发育相关基因的表达时序,维持细胞身份记忆,且多数成员在进化上高度保守。EZH1的特殊性在于其适应了终末分化细胞的表观遗传维持需求,与EZH2的增殖依赖性形成互补。
