FOSL1(FOS-like antigen 1)属于FOS基因家族,该家族还包括c-FOS、FOSB和FRA1等成员,这些基因编码的蛋白质均为转录因子,能与JUN家族蛋白结合形成AP-1(激活蛋白-1)转录复合物,调控细胞增殖、分化、凋亡及应激反应等关键生物学过程。FOSL1编码的蛋白FRA1(FOS-related antigen 1)通过AP-1复合物结合DNA特定序列,激活或抑制下游靶基因表达,参与细胞周期调控、上皮-间质转化(EMT)及炎症反应。其主要作用位点为细胞核,通过磷酸化等翻译后修饰调节其稳定性与活性。突变可能导致FOSL1功能异常,例如丧失DNA结合能力或影响AP-1复合物形成,进而干扰相关信号通路(如MAPK/ERK),与肿瘤发生发展密切相关,尤其在乳腺癌、肺癌和结直肠癌中高表达,促进侵袭转移。FOSL1过表达会增强EMT过程,上调基质金属蛋白酶(MMPs)等基因,加速肿瘤转移;还可能通过激活促生存信号导致化疗耐药。反之,降低表达可抑制肿瘤细胞迁移,但可能影响正常组织修复功能。FOSL1与炎症性疾病(如关节炎)也有关联,其过表达会加剧炎症因子(如IL-6、TNF-α)释放。FOS家族共性为:均含亮氨酸拉链结构域(介导蛋白二聚化),受生长因子或应激刺激快速诱导表达,且多为瞬时表达(但FOSL1稳定性较高)。需要注意的是,"FRA1"为FOSL1蛋白的常用别名,中文文献可能混用,需结合上下文区分。

The Fos gene family consists of 4 members: FOS, FOSB, FOSL1, and FOSL2. These genes encode leucine zipper proteins that can dimerize with proteins of the JUN family, thereby forming the transcription factor complex AP-1. As such, the FOS proteins have been implicated as regulators of cell proliferation, differentiation, and transformation.
