KDR(激酶插入结构域受体)也称为VEGFR2(血管内皮生长因子受体2),是血管内皮生长因子受体家族(VEGFR家族)的重要成员,该家族还包括VEGFR1(FLT1)和VEGFR3(FLT4)。KDR主要表达于血管内皮细胞表面,是VEGF信号通路中的关键受体,负责调控血管生成(angiogenesis)和血管通透性。当VEGF配体(如VEGF-A)结合KDR后,会触发受体二聚化和自磷酸化,激活下游信号通路(如PI3K-AKT和RAS-MAPK),促进内皮细胞增殖、迁移和存活,这对胚胎发育、伤口愈合和女性生殖周期等生理过程至关重要。KDR的功能异常与多种疾病密切相关,例如KDR突变或表达异常可能导致血管发育缺陷,而过度激活则促进肿瘤血管生成,为癌细胞提供营养和氧气,因此KDR是抗肿瘤药物(如贝伐珠单抗)的重要靶点。KDR的过表达会增强血管生成,可能加速肿瘤生长或糖尿病视网膜病变等血管增生性疾病;而降低表达则可能抑制血管形成,影响组织修复或导致缺血性疾病。VEGFR家族的共性在于均含有胞外免疫球蛋白样结构域、跨膜区和胞内酪氨酸激酶结构域,主要参与调控血管和淋巴管系统的发育与功能。研究表明KDR单核苷酸多态性(SNP)可能影响个体对心血管疾病或癌症的易感性,而某些激活性突变(如R1032Q)可能导致遗传性血管疾病。此外,KDR与其他基因如HIF-1α存在交叉调控,在低氧环境下HIF-1α可诱导VEGF表达进而激活KDR信号。由于其在血管生成中的核心作用,KDR已成为癌症、年龄相关性黄斑变性等疾病治疗的重要研究靶点。

Vascular endothelial growth factor (VEGF) is a major growth factor for endothelial cells. This gene encodes one of the two receptors of the VEGF. This receptor, known as kinase insert domain receptor, is a type III receptor tyrosine kinase. It functions as the main mediator of VEGF-induced endothelial proliferation, survival, migration, tubular morphogenesis and sprouting. The signalling and trafficking of this receptor are regulated by multiple factors, including Rab GTPase, P2Y purine nucleotide receptor, integrin alphaVbeta3, T-cell protein tyrosine phosphatase, etc.. Mutations of this gene are implicated in infantile capillary hemangiomas. [provided by RefSeq, May 2009]
