MYOG(Myogenin)是一种关键的肌源性调节因子(MRF),属于生肌调节因子(MRF)基因家族,该家族还包括MYOD1、MYF5和MRF4。这些基因在骨骼肌的发育和分化过程中起核心作用,通过调控下游肌特异性基因的表达来促进肌细胞的形成。MYOG主要在成肌细胞分化为成熟肌管阶段表达,是肌肉生成(myogenesis)的晚期标志物。其表达产物Myogenin是一种碱性螺旋-环-螺旋(bHLH)转录因子,通过结合E-box序列(CANNTG)激活肌肉特异性基因如肌球蛋白重链(MyHC)和肌酸激酶(CK)的转录。MYOG的功能缺失突变会导致严重的肌肉发育缺陷,如小鼠模型中表现为新生儿致死性肌无力,而人类中与先天性肌病相关。相反,MYOG的过表达可加速肌细胞分化,但若在非肌细胞中异位表达,可能诱导部分肌源性程序。该基因与肌肉萎缩性疾病(如杜氏肌营养不良)和某些肌肉肿瘤相关,其表达水平常作为肌肉再生能力的指标。在MRF家族中,MYOG的特点是其分化阶段特异性,而MYF5和MYOD1更早参与肌祖细胞决定,MRF4则在维持成熟肌纤维中起作用。MYOG表达受多种信号通路(如Wnt、Notch)和表观遗传调控,其下调会导致肌分化阻滞,而过表达可能抑制增殖相关基因,推动细胞周期退出。此外,MYOG与其他肌增强因子(如MEF2)协同作用,共同构建肌肉发育的转录网络。
