PARD3(Partitioning Defective 3)是PAR(Partitioning Defective)基因家族的重要成员,该家族还包括PARD1、PARD6等,主要参与细胞极性(cell polarity)的建立与维持,这是细胞在特定空间方向上形成结构和功能不对称性的过程。PARD3编码的蛋白质作为支架蛋白(scaffold protein),通过与PARD6、aPKC(非典型蛋白激酶C)等形成复合物(PAR复合物),调控细胞极性的关键信号通路,尤其在细胞紧密连接(tight junction)的形成、神经元轴突分化及上皮细胞极性中发挥核心作用。其主要作用位点包括细胞膜(如上皮细胞的顶侧膜)和神经元生长锥(growth cone)。PARD3突变可能导致其功能丧失,破坏细胞极性,进而引发多种疾病。例如,在癌症中,PARD3的缺失或突变与肿瘤转移相关,因其无法维持上皮细胞的粘附性,促进上皮-间质转化(EMT);在神经系统疾病中,PARD3异常可能影响神经元迁移或突触形成,与自闭症、精神分裂症等神经发育障碍有关。若PARD3过表达,可能增强细胞极性并抑制肿瘤侵袭,但异常高表达也可能干扰正常组织发育;而表达降低时,会削弱细胞间连接,导致屏障功能受损(如肠上皮通透性增加)或促进肿瘤恶性进展。PARD3基因家族的共性在于均含PDZ结构域(一种蛋白质相互作用模块),通过介导蛋白互作调控细胞极性相关通路。目前“PARD3”的中文译名“分区缺陷蛋白3”或“极性蛋白PAR3”均被使用,但后者更强调其功能特性。
