PLK4（Polo样激酶4，Polo-like kinase 4）是一种丝氨酸/苏氨酸蛋白激酶，属于Polo样激酶（PLK）家族，该家族成员（PLK1-5）均含有一个保守的Polo-box结构域，负责底物识别和亚细胞定位。PLK4是中心体复制的关键调控因子，通过磷酸化下游靶蛋白（如STIL、CEP152）确保中心体正确复制，维持基因组稳定性。其表达具有严格的剂量依赖性，轻微过表达即可诱导中心体过度复制，导致多极纺锤体形成和染色体错误分离，与多种癌症（如乳腺癌、肝癌）的发生相关；而PLK4缺失或功能丧失性突变会导致中心体缺失，引发有丝分裂缺陷、细胞凋亡或基因组不稳定性。PLK4在睾丸中高表达，其突变可能引起男性不育。该基因的异常表达还影响其他细胞周期调控基因（如p53、Cyclin B1）的功能。PLK家族共性包括：通过Polo-box结构域介导蛋白质相互作用，参与细胞周期调控（尤其是有丝分裂）、DNA损伤修复和中心体动态。目前针对PLK4的抑制剂（如CFI-400945）正在临床试验中用于癌症治疗。术语解释：中心体（centrosome）是细胞内的微管组织中心，参与纺锤体形成；丝氨酸/苏氨酸蛋白激酶（serine/threonine kinase）是一类将磷酸基团转移到特定丝氨酸或苏氨酸残基上的酶。
This gene encodes a member of the polo family of serine/threonine protein kinases. The protein localizes to centrioles, complex microtubule-based structures found in centrosomes, and regulates centriole duplication during the cell cycle.
