SYK(脾酪氨酸激酶,spleen tyrosine kinase)是一种非受体型酪氨酸激酶,属于SYK家族(包括SYK和ZAP-70两个成员),主要在造血细胞(如B细胞、T细胞、巨噬细胞等)中表达,参与免疫受体信号传导。其生物学功能是通过识别免疫受体(如BCR、Fc受体等)的ITAM基序(免疫受体酪氨酸激活基序),在磷酸化后激活下游信号通路(如PI3K-AKT、MAPK、NF-κB等),调控免疫细胞的增殖、分化、存活及炎症反应。SYK的特点是其双SH2结构域可同时结合两个磷酸化ITAM,从而放大信号。突变可能引起功能异常,例如功能获得性突变与自身免疫疾病或血液肿瘤(如弥漫大B细胞淋巴瘤)相关,而功能丧失性突变可能导致免疫缺陷。SYK过表达会过度激活免疫信号,与类风湿性关节炎、过敏性疾病及某些白血病相关;而降低表达可能抑制免疫反应,在自身免疫病治疗中具有潜力(如SYK抑制剂用于类风湿性关节炎)。SYK家族共性包括依赖SH2结构域识别磷酸化酪氨酸、参与免疫细胞信号转导及T/B细胞发育。其作用位点集中在免疫突触(如B细胞与抗原接触部位)和炎症病灶。专业术语解释:ITAM(免疫受体酪氨酸激活基序)是含特定酪氨酸的蛋白序列,磷酸化后招募SYK等激酶;SH2结构域是一种能结合磷酸化酪氨酸的蛋白模块。目前中文术语"脾酪氨酸激酶"为直译,临床常直接使用"SYK"。
This gene encodes a member of the family of non-receptor type Tyr protein kinases. This protein is widely expressed in hematopoietic cells and is involved in coupling activated immunoreceptors to downstream signaling events that mediate diverse cellular responses, including proliferation, differentiation, and phagocytosis. It is thought to be a modulator of epithelial cell growth and a potential tumour suppressor in human breast carcinomas.
