TSHR(甲状腺刺激激素受体,Thyroid Stimulating Hormone Receptor)是一种位于细胞膜上的G蛋白偶联受体(GPCR),属于糖蛋白激素受体家族。该基因主要在甲状腺滤泡细胞表面表达,负责结合垂体分泌的甲状腺刺激激素(TSH),激活下游信号通路(如cAMP/PKA和PLC/PKC途径),调控甲状腺激素(T3/T4)的合成与分泌,对机体代谢、生长发育和能量平衡至关重要。TSHR的突变可能导致功能异常:激活突变(如M453T)引发甲状腺功能亢进或毒性甲状腺腺瘤,而失活突变(如P162A)则导致先天性甲状腺功能减退。该基因与格雷夫斯病(Graves' disease,一种自身免疫性甲亢)密切相关,患者体内产生的TSHR刺激性抗体会持续激活受体。TSHR过表达可能引发甲状腺细胞过度增殖(如甲状腺癌),而表达降低则导致甲状腺激素不足(如呆小症)。其基因家族(糖蛋白激素受体家族)还包括LHCGR(黄体生成素受体)和FSHR(卵泡刺激素受体),共性为通过结合糖蛋白激素激活G蛋白信号,调控生殖与内分泌功能。专业术语解释:G蛋白偶联受体(GPCR)是一类跨膜蛋白,通过G蛋白传递细胞外信号;cAMP/PKA和PLC/PKC是细胞内信号转导通路,分别依赖环磷酸腺苷和磷脂酶C传递信息。

The protein encoded by this gene is a membrane protein and a major controller of thyroid cell metabolism. The encoded protein is a receptor for thyrothropin and thyrostimulin, and its activity is mediated by adenylate cyclase. Defects in this gene are a cause of several types of hyperthyroidism.
