USP28(泛素特异性蛋白酶28)属于去泛素化酶(DUBs)家族中的USP亚家族,该家族以含有高度保守的泛素特异性蛋白酶结构域为特征,主要功能是通过切除底物蛋白上的泛素链来逆转泛素化修饰,从而调控蛋白质稳定性、定位及功能。USP28在细胞周期调控、DNA损伤修复及癌症发生中起关键作用,其典型作用位点包括癌蛋白c-Myc和p53等,通过去泛素化稳定这些蛋白,促进细胞增殖或凋亡。突变可能导致USP28酶活性丧失或异常,例如功能缺失突变会削弱对c-Myc的稳定作用,抑制肿瘤生长,而功能获得性突变可能过度激活致癌通路。USP28与多种癌症(如结直肠癌、乳腺癌)密切相关,其过表达通常通过增强c-Myc和Notch等促癌蛋白的稳定性来驱动肿瘤进展,而敲低表达则可能诱导细胞周期停滞或凋亡。该基因还参与调控Wnt/β-catenin等信号通路,过表达会协同激活下游促增殖基因,而表达不足可能抑制通路活性。USP家族共性包括依赖半胱氨酸-组氨酸催化二元体(Cys-His box)水解泛素链,并普遍参与蛋白质稳态、炎症及表观遗传调控。目前“去泛素化酶”中文译名存在争议(原英文Deubiquitinating enzymes),部分文献也使用“去泛素酶”。
