WNT8A(Wingless-type MMTV integration site family, member 8A)属于WNT基因家族,该家族编码一组分泌型糖蛋白,在胚胎发育、细胞增殖、分化和组织稳态中发挥关键作用。WNT8A主要参与经典WNT/β-catenin信号通路(一种调控基因表达的细胞通讯机制),通过结合细胞膜上的Frizzled受体和LRP5/6共受体,激活下游信号传导,影响靶基因表达。其表达产物WNT8A蛋白在脊椎动物胚胎发育中尤为重要,尤其在神经管形成(胚胎中枢神经系统的早期结构)、体节分节(胚胎中轴骨骼和肌肉的前体组织)及肢体发育中起调控作用。该基因的突变可能导致信号通路异常,例如功能丧失突变会破坏胚胎形态发生,造成神经管缺陷(如脊柱裂)或肢体畸形;而功能获得性突变可能过度激活β-catenin,与某些癌症(如结直肠癌)的发生相关。WNT8A过表达会异常激活下游靶基因(如MYC、CCND1等促癌基因),促进细胞过度增殖;反之,表达降低可能导致发育障碍,如小鼠模型中Wnt8a缺失会引发中轴骨骼缺陷。WNT基因家族的共性包括:均编码富含半胱氨酸的分泌蛋白,通过自分泌或旁分泌方式(细胞自身或邻近细胞的信号传递)调控保守的信号通路,且多数成员在进化中高度保守。目前研究还发现WNT8A在成体组织中表达有限,但在某些肿瘤中异常激活,提示其可能成为癌症治疗的潜在靶点。专业术语解释:经典WNT通路指依赖β-catenin进入细胞核调控转录的路径;自分泌/旁分泌指细胞分泌信号分子作用于自身或附近细胞。

The WNT gene family consists of structurally related genes which encode secreted signaling proteins. These proteins have been implicated in oncogenesis and in several developmental processes, including regulation of cell fate and patterning during embryogenesis. This gene is a member of the WNT gene family, and may be implicated in development of early embryos as well as germ cell tumors.
