ZAP70(Zeta-chain-associated protein kinase 70)是一种主要在T细胞和自然杀伤(NK)细胞中表达的酪氨酸激酶,属于SYK家族(Spleen tyrosine kinase family)的成员。该基因家族的共性是其编码的蛋白均含有两个SH2结构域(Src homology 2 domains,负责结合磷酸化酪氨酸残基)和一个激酶结构域(kinase domain,具有催化活性),在免疫受体信号传导中起核心作用。ZAP70的主要生物学功能是参与T细胞受体(TCR)的信号转导:当TCR与抗原结合后,ZAP70被招募至激活的CD3链磷酸化位点,通过其SH2结构域结合磷酸化的免疫受体酪氨酸激活基序(ITAMs),随后自身磷酸化并激活下游信号分子如LAT和SLP-76,最终导致T细胞活化、增殖及细胞因子分泌。该基因的作用位点集中在免疫突触(T细胞与抗原呈递细胞接触形成的特化结构)内。ZAP70突变会导致严重联合免疫缺陷病(SCID),表现为T细胞功能缺失而B细胞数量正常但功能异常(因B细胞依赖T细胞辅助),患者易发生反复感染。在慢性淋巴细胞白血病(CLL)中,ZAP70表达水平是预后指标,其过表达(通常通过甲基化调控异常导致)会增强BCR信号通路活性,促进恶性B细胞存活和增殖;而在类风湿性关节炎中,ZAP70过表达可能加剧自身免疫反应。若该基因表达降低(如基因沉默或药物抑制),会削弱T细胞激活,可用于治疗自身免疫疾病或器官移植排斥,但可能增加感染风险。ZAP70与SYK家族另一成员SYK具有部分功能冗余,但在淋巴细胞中ZAP70占主导地位。其表达严格受发育调控,胸腺细胞成熟过程中ZAP70替代SYK成为主要信号分子。最新研究发现ZAP70还参与调节T细胞代谢重编程和记忆T细胞形成,其激酶非依赖功能(如支架作用)也逐渐被揭示。

This gene encodes an enzyme belonging to the protein tyrosine kinase family, and it plays a role in T-cell development and lymphocyte activation. This enzyme, which is phosphorylated on tyrosine residues upon T-cell antigen receptor (TCR) stimulation, functions in the initial step of TCR-mediated signal transduction in combination with the Src family kinases, Lck and Fyn. This enzyme is also essential for thymocyte development. Mutations in this gene cause selective T-cell defect, a severe combined immunodeficiency disease characterized by a selective absence of CD8-positive T-cells.
