RPRM(Reprimo)是一个重要的抑癌基因,属于Reprimo基因家族,该家族成员在细胞周期调控中发挥关键作用,尤其是参与G2/M期检查点的调控。RPRM基因编码的蛋白通过与细胞周期蛋白依赖性激酶(CDK)相互作用,抑制CDK活性,从而阻止细胞周期进程,确保DNA损伤修复完成后再进入有丝分裂。其主要作用位点在细胞质和细胞核,通过表观遗传调控(如DNA甲基化)影响其表达。RPRM的突变或表观遗传沉默可能导致其功能丧失,使细胞周期检查点失效,增加基因组不稳定性,进而促进肿瘤发生。研究发现,RPRM在多种癌症(如胃癌、乳腺癌和结直肠癌)中表达降低或缺失,与肿瘤进展和不良预后相关。若RPRM过表达,可增强细胞周期阻滞,抑制肿瘤细胞增殖;而降低表达则可能导致细胞周期失控,促进肿瘤发展。此外,RPRM与p53信号通路密切相关,p53可激活RPRM转录,进一步强化其抑癌功能。Reprimo基因家族的共性在于它们均参与细胞周期负调控,尤其在DNA损伤应答中协调细胞周期停滞,维持基因组稳定性。RPRM还与其他基因如CDK1、CDC25等相互作用,共同调控G2/M期转换。其表达异常不仅影响自身功能,还可能扰乱整个细胞周期网络,导致疾病发生。
