TAF5L, or TATA-box binding protein associated factor 5-like, is a member of the TAF gene family that encodes essential subunits of the TFIID general transcription factor complex, which serves as the primary platform for initiating RNA polymerase II-mediated transcription. As a core component of this multi-protein assembly, TAF5L utilizes a conserved histone fold domain to form stable heterodimers with other TAF proteins, thereby facilitating the structural integrity of TFIID and enabling the precise recognition of core promoter elements, such as the TATA box, to recruit the RNA polymerase II machinery to transcription start sites. While the TAF family broadly regulates basal transcriptional mechanisms, TAF5L exhibits a distinct biological profile characterized by high expression in the testis, where it plays a critical role in spermatogenesis and the differentiation of germ cells.
